The combination of Glimepiride and Pioglitazone is typically used to treat type 2 diabetes mellitus when diet, exercise, and single-drug therapy (such as metformin or sulfonylurea) do not provide adequate glycemic control.
Common side effects of this combination may include weight gain, edema (fluid retention), headache, dizziness, and hypoglycemia (low blood sugar). Less common but more serious side effects may include liver problems, heart failure, and an increased risk of bladder cancer.
This combination is contraindicated in patients with a history of hypersensitivity to glimepiride, pioglitazone, or any other component of the formulation. It should also be avoided in patients with severe heart failure, liver disease, or diabetic ketoacidosis.
Glimepiride is a sulfonylurea that stimulates the release of insulin from pancreatic beta cells, thereby lowering blood sugar levels. Pioglitazone is a thiazolidinedione that improves insulin sensitivity in muscle and adipose tissue and reduces hepatic glucose production.
Patients taking this combination should be monitored regularly for signs and symptoms of heart failure, liver dysfunction, and bladder cancer. It may also be necessary to adjust the dosage of concomitant medications, such as insulin or other oral antidiabetic agents, to avoid hypoglycemia. Pregnant and lactating women should use this combination with caution, as its safety in these populations has not been established.
