Rucaparib is a medication used to treat certain types of cancer. It is in a class of medications called poly (ADP-ribose) polymerase (PARP) inhibitors. PARP is an enzyme that plays an essential role in DNA repair. Rucaparib works by blocking PARP, which can lead to cancer cell death.

    • Treat certain types of ovarian cancer, fallopian tube cancer, and primary peritoneal cancer in people with a specific gene who have received treatment with at least two other chemotherapy treatments.
    • Treat certain types of prostate cancer that has spread to other areas in the body in people with a specific gene who have received other treatments.

Mechanism of Action

Rucaparib is a PARP inhibitor. PARP is an enzyme that helps cells repair damaged DNA. Rucaparib works by blocking PARP, which can lead to cancer cell death.

Cancer cells often have DNA damage that makes them more likely to die. Rucaparib can help these cells die by preventing them from repairing their damaged DNA.

Rucaparib is most effective in cancer cells that have mutations in the BRCA1 or BRCA2 genes. These genes are involved in DNA repair, so mutations in these genes make cells more dependent on PARP for DNA repair.
