Sildenafil is used in the treatment of Erectile Dysfunction . Erectile dysfunction is the inability to achieve or maintain an erection long enough to perform sexual intercourse.

Side Effects of Power Vegra 100mg Tab 4s

Common Side effects of Sildenafil :

1. Headache,
2. Flushing,
3. Back pain and muscle pain
4. Respiratory tract infection,
5. Runny nose
6. Angina pectoris, AV block,
7. Migraine, syncope,
8. Tachycardia, and postural hypotension
9. Tremor, depression,
10. Herpes simplex,
11. Skin ulcer,
12. Oesophagitis,
13. Hypoglycaemic reaction

Drug Interactions

1. Sildenafil excretion from the body is reduced by some medicines like erythromycin and cimetadine (Inhibitors of CYP3A4 enzyme in liver) so its effect or side effects may last longer in patients taking these medicines.
2. Rifampicin may decrease the plasma concentrations of sildenafil.
3. Symptomatic hypotension may occur when sildenafil is used along with beta lockers.
4. Plasma concentrations of sildenafil is increased by ritonavir.
5. Potentially Fatal: Sildenafil may increase hypotensive effects of of organic nitrates and nicorandil.

Contraindications

1. Sildenafil should not be used in patients who show hypersensitivity to this medicine.
2. Patients concurrently or intermittently using organic nitrates in any form.

Mechanism of Action

Sildenafil inhibits phosphodiesterase type-5 (PDE5) which is responsible for cGMP degradation in the corpus cavernosum. Inhibition of PDE5 increases cGMP levels in the corpus cavernosum which results in smooth muscle relaxation and inflow of blood to the penis ( corpus cavernosum.)

Special Precautions

Caution when used in patients with anatomical deformation of penis or conditions that may predispose them to priapism (e.g. sickle cell anaemia, myeloma, or leukaemia). Mild, transient, dose-related impairment of colour discrimination (blue/green) may occur. Hepatic or severe renal impairment, bleeding disorders, active peptic ulceration, hypotension, recent history of stroke, MI, arrhythmias, unstable angina, heart failure or retinal disorders. May cause sudden loss or decrease in hearing.
